My rear passenger power lock actuator is not functioning, and makes a whirring noise when I hit the power locks. I picked up one today in the junkyard out of a 99.

I don't really want to tear apart the door, just to check to see if it works correctly.

Could it be as simple as hooking it up to a 12V power source?
Could it be as simple as pos/neg to lock, and neg/pos to unlock (or vice-versa)?

Yes, that simple.
