Strange noise while driving

Hey everybody - new to this forum so I hope that I am posting this in the correct spot.
I have a 96 Explorer 4WD v6 4L and I have recently aquired a strange noise while driving that I am at a loss as to what it surely is. When I start driving its makes a thudding or "duh" noise (its rather difficult to describe and thats the best that I can do unfortunately). I first thought that it was something to do with my tires but the speed of the vehicle/tires does not match the noise interval at all - it is too slow to be the tires. It only lasts for about 5-8 minutes while first starting to drive then goes away, it is only when I step on the gas - and when I switch to 4wd it goes away.
I have had tranny problems before in a previous vehicle the Taurus (oh the Taurus Tranny :) ) and it feels like a very very slight tranny buck - like going over a small bump in the road. Thinking this I flushed and changed the filter and fluid in the tranny - no change - Is this truly a tranny demon or is it some other gremlin in my Explorer?

Update : So I checked the level in my Transfer Case after driving it with my Brother so that he could hear the noise, the Transfer Case was low on fluid. After discovering this I filled it up. The noise is now less but it is still there and it appears that it is getting more constant - lasting the entire drive not just 5 - 8 minutes at the start.
Any thoughts?
