Trailer towing with 5R55E

2000 V6 with 5R55e auto Tranny

I need to tow a trailer about 200 miles and the total weight will be 4-5,000 lbs for the trailer.

Do you think the trans can handle it, or should I make other arrangements?

I have no trans cooler, and I plan on going 50 MPH, and with OD off.

If this will be a one-time event, you s/b okay. Make sure you have a hitch capable of towing 5,000 lbs. As you say, don't run in O/D. If you haven't had your trans fluid changed in a while, that wouldn't hurt either.
