i don't what that noise is but if you run out of diff fluid the entire third member will expand after it gets to a certain temperature although each piece of the differential will expand at a different rate but once one piece starts expanding it will wear on the other parts. granted it won't 'blow up' as soon as you run out, but the fluid is designed for lubrication and cooling so once the fluid is gone you will get premature stress and fatigue on all the parts that need the properties of the fluid to function correctly
EDIT: i know the rear differential will make a noise like that when the pinion isn't securely tightend, but i don't have any experience with front differentials.
 


















ya it could be a number of things: loose pinion, chipped axle spline, chipped ring gear, chipped pinion, etc...either way it won't be cheap to fix
